Steven Mullaney struck a century from 55 deliveries - Nottinghamshire's fastest ever in the County Championship - before late wickets handed the Green and Golds the chance to push for a final-day win.
Resuming on 6/0 and leading overall by 226 runs, Luke Fletcher - as an opener - hit his second fifty of the game as the lead passed 350.
Mullaney then hammered a fifty from 32 balls before accelerating even more, reaching his ton just before the visitors declared on 295/5.
That set Middlesex a nominal 516 to win, and after Fletcher and James Pattinson picked up three evening session wickets, the Green and Golds will resume requiring seven further scalps for a third successive Championship victory.
