Tom Moores scored 34, but nobody else passed 20, as the hosts were all out for 165 in reply to the visitors’ 328 all out.
Left-arm spinner Callum Parkinson, a close season signing from Derbyshire, took four wickets for eight runs. Former Worcestershire paceman Richard Jones ended with figures of 3-36.
The lead had been extended to 334 by the close; Matt Milnes, Ben Kitt and Matt Carter sharing the wickets as Leicestershire reached 171-4.
