Mark Wagh and Matt Wood hit season-best scores as Notts declared on 558-7 on day three at Hove.
Play closed with Sussex 212 runs behind having reached seven without loss in their second innings.
Wagh was out lbw after hitting 9 fours and a six on his way to 98 with Wagh recording 128, his highest score for 13 months.
“The three days have gone to plan and the worst case scenario is that we leave with a draw and maximum bonus points,” said Notts Director of Cricket Mick Newell.
“Mark Wagh played very well and it was an excellent knock from Matt Wood and we’ve created an opportunity to win the game.
“It’s going to be a hard-fought final day and winning the game will be a battle but we’re in with a shout.”
Ashwell Prince was caught at slip for 30 and Chris Read (38), Paul Franks (25) and Rob Ferley (26) made solid contributions.
